Former Sen. George Allen enters the primary election favored over three lesser-known Republicans as he tries to win back the Senate seat he lost six years ago.
Polls are open from 6 a.m. to 7 p.m. Tuesday.
Allen's challengers are tea party leader Jamie Radtke, state legislator Bob Marshall and minister E.W. Jackson.
A Washington Post poll from last month showed that more than six out of 10 likely GOP primary voters favored the former governor who lost his 2006 Senate re-election to Democrat Jim Webb.
The winner faces Democratic former Gov. Tim Kaine in a race that could determine Senate partisan control.
Six U.S. House nominations will also be decided. Challenged incumbents include Democrat Jim Moran and Republican Reps. Randy Forbes, Bob Goodlatte and House Majority Leader Eric Cantor.
